Contraindications

Do not take compounded semaglutide if you:

  • Have a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN2).
  • Are allergic to GLP-1 medications (Adlyxin®, Byetta®, Bydureon®, Ozempic®, Rybelsus®, Trulicity®, Victoza®, Wegovy®) or their inactive ingredients: di-sodium hydrogen phosphate dihydrate, sodium chloride, benzyl alcohol, hydrochloric acid, sodium hydroxide pellets, and water.
  • Have diabetes (Type 1 or 2)
  • Have been diagnosed with pancreatitis or history of pancreatitis
  • Have gastroparesis or have a history of gastroparesis
  • Have a history of suicidal attempts or active suicidal ideation
  • Have hypoglycemia
  • Have acute kidney disease
  • Have acute gallbladder disease
  • Have hypersensitivity reactions
  • Are planning to conceive within the next 2 months
  • Are pregnant
  • Have diabetic retinopathy
  • Are breastfeeding
  • Less than 18 years old

Before taking compounded semaglutide, tell your doctor if you are pregnant, breastfeeding, or planning on conceiving in the next two years, or if you have any of the below conditions:

  • Diabetes (Type 1 or 2)
  • Pancreatitis
  • Diabetic Retinopathy
  • Hypoglycemia
  • Gallbladder disease
  • Kidney disease or acute kidney injury
  • Hypersensitive reactions
  • Depression or suicidal ideation

Possible Side Effects

The most common side effects of compounded semaglutide include nausea, diarrhea, vomiting, constipation, stomach (abdomen) pain, headache, tiredness (fatigue), upset stomach, dizziness, feeling bloated, belching, low blood sugar in people with type 2 diabetes, gas, stomach flu, heartburn, and runny nose or sore throat.

Compounded semaglutide may also cause:

  • Inflammation of the pancreas (pancreatitis)
  • Gallbladder problems
  • Increased risk of low blood sugar (hypoglycemia), especially for those who also take medicines for diabetes such as insulin or sulfonylureas
  • Kidney problems (kidney failure)
  • Serious allergic reactions, including swelling of the face, lips, tongue, or throat; problems breathing or swallowing; severe rash or itching; fainting or feeling dizzy; or very rapid heartbeat
  • Change in vision in people with type 2 diabetes
  • Increased heart rate
  • Depression or thoughts of suicide
  • Food or liquid getting into the lungs during surgery or other procedures that use anesthesia or deep sleepiness (deep sedation)

Warnings

In rodents, semaglutide causes dose-dependent and treatment-duration-dependent thyroid C-cell tumors at clinically relevant exposures. It is unknown whether semaglutide causes thyroid C-cell tumors, including medullary thyroid carcinoma (MTC), in humans as human relevance of semaglutide-induced rodent thyroid C-cell tumors has not been determined.

Semaglutide is contraindicated in patients with a personal or family history of MTC or in patients with Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).
